Faculty Profile

I am working as an Associate Professor in the Department of Pharmaceutical Engineering & Technology at the Indian Institute of Technology (BHU), Varanasi since November 2024. I obtained my Ph.D. degree in Pharmaceutical Sciences from the University Institute of Pharmaceutical Sciences, Panjab University, Chandigarh, India in 2014 on the topic "Synthesis and study on azasteroids as 5α-reductase inhibitors." Soon after the Ph.D. thesis defense in April 2014, I moved to the Centre for Alzheimer Research, Department of Neurobiology, Care Sciences and Society, Karolinska Institutet, Stockholm, Sweden as a postdoctoral research associate, working with Prof. Taher Darreh-Shori and Prof. Bengt Winblad.

In June 2019, I accepted an Assistant Professor position at the Centre for Alzheimer Research, Karolinska Institutet, Stockholm, Sweden, and moved to India in November 2019.

I have also been awarded the Japan Society for the Promotion of Science summer fellowship to work in the Mizuguchi lab headed by Prof. Kenji Mizuguchi at NIBIOHN, Osaka, Japan during the summer of 2019. My main interests during my stay in Japan were to use Bioinformatics tools such as TargetMine developed by the Mizuguchi lab to discover novel drug targets using multi-omics data and AI/ML-based tools for pharmacokinetic properties prediction. My current research work is focused on the design, synthesis, and evaluation of potential therapeutics for the treatment of neurodegenerative diseases and to tackle antimicrobial resistance.

Our Research Projects

1. Design and Synthesis of Natural Product-Inspired Molecules to Target Protein Aggregation in Neurodegenerative Diseases

Protein aggregation plays a pivotal role in the progression of many neurodegenerative diseases, making it a critical drug target for therapeutic intervention. Our research focuses on the design and synthesis of small molecules inspired by natural products to target these aggregation processes in disorders like Alzheimer's and Parkinson's disease. We are specifically developing compounds that can modulate the aggregation of proteins such as amyloid-beta and tau in Alzheimer's disease, as well as alpha-synuclein in Parkinson's disease. By targeting these aggregation pathways, we aim to prevent or mitigate the toxic effects of protein accumulation that drive neurodegeneration.

2. Discovery and Development of Choline Acetyltransferase (ChAT) ligands for Early Diagnosis of Alzheimer's Disease

Alzheimer's disease and related dementias present significant challenges, making early diagnosis crucial for improving therapeutic outcomes. Growing scientific evidence identifies the cholinergic enzyme ChAT (choline acetyltransferase), essential for acetylcholine biosynthesis, as a promising in vivo target for developing ligands that could serve as probes for early diagnosis or therapeutic monitoring. Despite its potential, no imaging ligands for ChAT are currently available. Our research is focused on filling this gap by designing and developing novel ligands that can be utilized as probes for imaging, including PET tracers, to target ChAT. This multidisciplinary effort involves the computational design, synthesis of potential candidates, in vitro and in vivo studies. We aim to develop valuable tools for early detection and monitoring therapeutic responses in Alzheimer's and related dementias.

3. AI/ML-enabled Discovery and Development of Lead Compounds

AI and ML have revolutionized the discovery and development of lead compounds by significantly accelerating and refining the process. We utilized AI/ML for virtual screening of large chemical libraries, Deep Learning for predictive modeling of ADMET parameters, and Generative modeling for de novo drug design for a variety of diseases such as diabetes, antimicrobial resistance, cancer, and neurodegeneration. This approach enhances precision in drug design, facilitating the rapid development of effective lead molecules.
